ACT Policing said the accident has closed the Monaro Highway, between Williamsdale and the NSW border.
The crash, between a ute and truck, happened at about 3.30am.
The man who died was driving the ute and was the only person in that vehicle at the time.
"The driver of the semi-trailer did not sustain any injuries and is assisting police with the investigation," police said in a statement.
"The Monaro Highway, between Royalla and Williamsdale will remain closed to allow the Collision Investigation and Reconstruction Team to investigate the collision."
Police initially set up a diversion for light vehicles, through Williamsdale Road, Burra Road and Old Cooma Road, but trucks were are being forced to wait by the side of the road.
Police opened one lane to allow traffic to resume on Monday afternoon.
The crash left a large amount of debris on the road and the ute was severely damaged.
ACT Policing's Acting Sergeant Travis Lutz said it was a sad morning as police investigate the second fatality on ACT roads in one week and the seventh this year.
"The truck driver is receiving support from us," he said.
"There were a number of witnesses who are also receiving support. They arrived shortly after the incident occurred and notified police."
Acting Sergeant Lutz urged motorists to slow down and drive to conditions, be aware of fatigue and not drive while distracted.
Heavy fog significantly clouded visibility on the Monaro Highway near the crash site about 7am in the morning.
Acting Sergeant Lutz said the weather conditions would form part of the investigation.
He would not provide any details about the drivers or the direction the vehicles were travelling.
The death comes after one man was killed and two people were injured in a smash between a motorcycle and a car in Canberra on Friday.
Police said a 23-year-old man died when his motorbike and the BMW collided on Lady Denman Drive about 8.30am.
Friday's death coincided with the publication of the 2015 ACT Road Crash Report, which revealed about a third of the people who died or were injured on Canberra roads last year were cyclists, pedestrians and motorcyclists.
