The woman in her 60s was the driver of a vehicle that collided with a truck, about four kilometres north of Hall about 4.10pm.
NSW and ACT police and ambulance crews assisted in extracting the male driver from the truck before he was treated by paramedics and taken to Canberra Hospital.
At 6pm, the Barton Highway remained closed in both directions from Hall, with diversions expected to be in place for several hours.
Officers from the Monaro Local Area Command have established a crime scene in the area, with specialist forensic officers expected to examine the scene on Friday night.
All drivers are being urged to use the Federal Highway, Sutton Road and then Murrumbateman Road to and from Murrumbateman.
ACT Policing is assisting with traffic management and have warned motorists to expect significant delays.
NSW Police Media confirmed a report would be prepared for the coroner following the fatal collision.
Anyone who has not yet spoken to police about the collision or has information that could assist investigators is asked to contact Crime Stoppers on 1800 333 000.
The accident is the death latest on the Barton Highway after a 57-year-old motorcyclist was killed last month and a 27-year-old Canberra man died in February.
