The damage to Dean's rotator cuff is worse than first thought and he wasn't considered for Monday's match with Tasmania at Blundstone Arena.
Comets officials are resigned to Dean missing the following the two games against NSW at Manuka Oval and Queensland at the Gabba.
His absence is also going to test the depth of Weston Creek Molonglo as they aim to qualify for the finals of the Douglas Cup.
The Comets have made three changes to the side which lost to Western Australia.
Weston Creek fast bowler Joe Slater will make his Comets debut, Nathan McAndrew is recalled after playing for Sydney Thunder and batsman Henry Hunt returns from Australian under-19 commitments in Dubai.
Dean Solway, Josh Bennett and Mitchell Phelps are the players to miss out.
Jono Dean wasn't considered with his commitments with the Adelaide Strikers, who faced Sydney Thunder in Thursday night's Big Bash League semifinal.
Comets: Michael Spaseksi (c), Matt Condon, Mac Wright, Kerrod White, Tom Rogers, Andrew Harriott (wk), Henry Hunt, Blake Macdonald, Joe Slater, Nathan McAndrew, Cameron Suidgeest, Ben Taylor.
