South Coast was bundled out for just 68 before Andrew Stead (29) and captain Mark Bennett (29 not out) guided the hosts past the target with more than eight overs to spare.
Bennett believed the win would be a confidence boost heading into next weekend's switch to the two-day competition.
''It was a bit of an unknown game with none of us knowing any of the South Coast boys,'' Bennett said.
''We lost the toss and had to bowl first and bowled a lot better than we did yesterday. We took a few early wickets, which always helps, and kept taking wickets throughout the innings.''
ANU conceded 3-171 in its 67-run loss to Tuggeranong on Saturday, with Matt Floros doing most of the damage with an unbeaten 122.
ANU has wasted no time in turning its focus to the longer format of the game.
''It's going to be hard for us to go from playing one-dayers last week, Twenty20s this week and two-dayers next week,'' Bennett said.
''You just have to change your mindset a little bit.
''We've already started talking about it, there's a bit more patience required.''
Meanwhile, North Canberra Gungahlin capped a successful six-hour return road trip by knocking off defending champions Albury Wodonga by 41 runs.
North Canberra Gungahlin reached 5-160 from its 20 overs before restricting Albury Wodonga to 9-119.
Albury Wodonga (1-84 off 10 overs) bounced back to beat Ginninderra (8-80) by nine wickets.
The ACT under-19s (7-152) chased down Newcastle's total of 149 at Campbelltown with two balls to spare, while Weston Creek-Molonglo (140) also defeated Newcastle (9-113) by 27 runs.
