The driver of the car, a 55-year-old Deer Park woman, had been driving up Winnington Street and was attempting a U-turn at the intersection with the road.
It is believed she then lost control of the vehicle and struck the victim before crashing into a brick fence.
The 65-year-old woman died at the scene.
Police are investigating the cause of the crash.
The woman's death brings the road toll for the year up to 30, two fewer than at the same time in 2015.
It was the second death on Victoria's roads on Friday. A 27-year-old Gladstone Park motorcyclist died in hospital this morning following a serious collision in St Albans on Wednesday January 27.
