Wiasak outsprinted former winners Sarah Roy and Kimberley Wells, who was third, on day one of the nationals in Ballarat.
Wiasak is a two-time world champion in the individual pursuit.
Earlier on Wednesday, NSW rider Cameron Scott won the men's under-23 criterium.
Scott, who turns 20 on Thursday, was in a four-man breakaway and he outsprinted Dylan Sunderland for the win.
Rio Olympics team pursuit silver medallist Sam Welsford was four seconds behind in third.
Penrith rider Stephen Cuff and Melbourne's Sarah Gigante won the under-19 criteriums.
