A Federal Court judge on Friday rejected Michaela Banerji's third application for a stay on her dismissal.
The decision means Ms Banerji's job at the Immigration Department ends at close of business today.
The department sacked the public affairs officer after she tweeted comments that were critical of the government, the department and its spokesman, Sandi Logan.
Ms Banerji’s initial attempt to stay her dismissal was rejected by the Federal Circuit Court last month.
The department gave Ms Banerji a notice of termination, taking effect at close of business on Friday, after last month's judgment was made.
She then on Monday unsuccessfully applied for a stay on her sacking until her appeal was heard in the Federal Court next month.
Ms Banerji appealed the rejection of that application on Friday, seeking to keep her job until after the appeal.
Justice Peter Jacobson found there was no reason to overturn the previous decisions.
