The Toys 'R' Us store was among the 44 closed across the country last year, when 700 staff members lost their jobs nationwide.
The new Toymate store will see the creation of about 25 local jobs, according to Toymate's Danny Bloom.
"We are excited to become part of the Canberra community and we are looking to share our passion for toys," Mr Bloom said.
Capital Airport Group property head Richard Snow welcomed the announcement.
"Toymate will bring a new retail experience for Canberrans," Mr Snow said.
"Both kids and parents will love the in-store experience, and Toymate's customer service is exceptional. They pay attention to what customers want, including a massive range of brands, competitive pricing and even a dedicated parents' room in the store. We know our shoppers will love it."
Australian owned and operated, Toymate plans to open in Canberra before Christmas as it takes over the 1560-square-metre space.
"Toymate has an excellent reputation. It's a perfect fit for shoppers at Majura Park and the first stage in the redevelopment of that part of the centre that will eventually accommodate up to five new retailers," Mr Snow said.
The chain store offers a range of toys including robotics, crafts, games, outdoor toys and more. It will also have an area dedicated to babies and preschoolers and will include a parents' room.
Toymate currently has locations in New South Wales and Western Australia and a superstore on the Gold Coast.
