The man, an ACT resident, died at the scene after the car he was travelling in hit a tree on Belconnen Way at 4.20am on Wednesday.
It is the third fatal incident on ACT roads so far this year.
A female passenger was in the car at the time of the crash, and was taken to Canberra Hospital with minor injuries.
Acting Station Sergeant David Wills said there was significant damage to the driver's side of the car.
"The investigation is very early, and a team is putting together a picture of the circumstances leading to the collision, and looking to whether speed or other things that might have contributed," Acting Station Sergeant Wills said.
The victim's family has yet to be informed of the crash, police say.
Police are yet to speak to the female passenger, but said she was lucky to escape the crash with minor injuries.
"It's quite remarkable, the damage to the car was substantial, with injuries causing death, so for her to escape with relatively minor injuries is remarkable," Acting Station Sergeant Wills said.
Police issued a reminder to drivers to be safe on the roads in the wake of the fatal crash.
"Any death on the road is one death too many," Acting Station Sergeant Wills said.
A report will be prepared for the Coroner.
Anyone who may have witnessed the crash or has dash-cam footage has been urged to contact police on 131 444.
