Following weeks of tongue-in-cheek smear campaigns against his fellow nominees, Gleeson triumphed with the first Gold Logie win for the ABC in 40 years.
"Do I have any regrets? No, I got a result," Gleeson said.
"I'm like Scomo on election night, this is a victory for the quiet Australians.
"I can tell you this, I am not the most popular person on Australian television. Turns out I am just really good at manipulating the media to get people to do things for me.
"Which, if you think about it, is why we're all here. So maybe I do deserve this award."
Fellow nominees Amanda Keller, Rodger Corser, Sam Mac, Eve Morey, Waleed Aly and Costa Georgiadis were left empty-handed at the TV Week Logie Awards.
Landscaping guru Georgiadis had been on a roll, with his Gardening Australia show picking up two awards, for best presenter and most popular lifestyle program.
Gleeson was so sure he'd lose to his ABC colleague he admitted he'd been drinking heavily ever since Georgiadis took out best presenter.
Dylan Alcott, who recently won the quad wheelchair French Open title, won the Graham Kennedy award for most popular new talent on Sunday night.
His television breakthrough came as a co-host of the Invictus Games Tonight show and The Set.
English actress Jenna Coleman won the Silver Logie for best actress for her role in the ABC drama, The Cry, while Scott Ryan won Silver for gritty Foxtel drama Mr Inbetween.
The Stan series, Bloom, won two awards - most outstanding mini series or telemovie and most outstanding supporting actress for industry veteran Jacki Weaver.
The ABC's Mystery Road was a surprise winner of the most popular drama program, defeating high-rating commercial shows such as Home and Away and Doctor Doctor.
Kerry O'Brien from the 7.30 Report was given a standing ovation after he was inducted into the Logie Hall of Fame.
But the night's biggest winner was the ABC, beating out commercial rivals Seven and Nine, which failed to pick up a single win.
