A scratchy Warner, who made 94, rode his luck through the first two sessions, bowled by Ben Stokes on 17 but recalled when replays showed he had overstepped.
The opener was dropped in the slips by Rory Burns on 49 and then Haseeb Hameed's shy at the stumps from short-leg missed, Warner on 60 and short of his ground as he dropped his bat attempting to make his ground.
Playing and missing almost 20 times at deliveries from Mark Wood and Ollie Robinson combined, Warner's luck finally ran out early in the first session, tamely mistiming a drive to cover.
He and Labuschagne (74) put on 156 to see the hosts past England's first innings total of 147, Warner incredibly falling just one shy of his entire 2019 Ashes series aggregate.
It was a scratchy return to Test cricket in what was Warner's first red-ball innings since claiming player of the tournament honours in Australia's victorious Twenty20 World Cup campaign.
There were glimpses of that form though, Warner greeting Jack Leach into the attack with two straight sixes.
Labuschagne joined in with two of his own, before his commanding innings was quickly halted by a cut shot that ballooned to point.
The pair were left to rue their exits though as triple-figure eluded them and England kept their noses in the contest.
