It also won the guild's Excellence in Professional Innovation Award.
The Pharmacy of the Year award is regarded as "the undisputed benchmark for recognising excellence in the community pharmacy industry".
It aims to seek out "highly-motivated, innovative and adaptable pharmacy businesses that never sit still, and strive to provide further health care to support their community".
The awards were presented by Pharmacy Guild of Australia national president Trent Twomey at the Australian Pharmacy Professional conference on the Gold Coast on Thursday.
Cooleman Court Pharmacy pharmacists and owners Brad Butt and Brooke Veasey said "the team was overwhelmed but thrilled to be recognised by their industry as pharmacy of the year".
"Although it is great to win such an auspicious award, it is making a difference to our local community that keeps our team coming to work each day," they said.
"That said, we are so pleased that the team won this award and get the recognition that comes along with it."
Cooleman Court Pharmacy was renovated in 2018 and had multiple clinic rooms built as a core part of the development. These clinic rooms not only run the now ubiquitous pharmacy vaccination and nursing services, but also serve as a base to the Men's Health Downunder clinics established by Brad, which assist men with post-prostate surgery rehabilitation and related issues.
"My goal with the pharmacy was to invest in its people and establish a great working environment with a strong emphasis on delivering a wide range of professional services to the community," Mr Butt said.
Ms Veasey started as an intern at the pharmacy because "it was known as being one of the most innovative pharmacies in the region".
"Since completing my internship with Brad, I have seen first-hand the community and patient-first attitude displayed by all of the team and was last year extremely proud to join as a co-owner of the pharmacy," she said.
Judging panel member Nick Panayiaris said the Cooleman Court Pharmacy stood out among the 200 nominations "in how it has succeeded in developing into a community health hub".
"The pharmacy downsized and made the conscious decision to rationalise non-pharmacy lines from their operations and to focus on healthcare services and information," he said.
"They also actively empowered employees to develop projects and ideas, taking ownership and responsibility in driving new opportunities in their community pharmacy".
