Bars, restaurants, clubs and hotels are running the game in the early afternoon, alongside other Anzac Day events.
The Canberra Services Club and the Belconnen Labor Club are holding two-up games for the first time since the pandemic began.
Canberra Services Club president Michael Kinniburgh said getting back to annual two-up competitions was important to the club.
"We have been having games since 1939, so it's a huge deal," he said.
"We haven't had any games for the past two years, as we didn't want to put any of our older patrons at risk.
"It's going to be absolutely amazing. It's always such a great day."
Belconnen's Canberra Labor Club is also hosting two-up for the first time since 2019. The games start at noon, and the Anzac Day footy will be streamed during the day.
Olims Bar & Bistro, located in Mercure Canberra just 600 metres from the Australian War Memorial, is hosting two-up, breakfast and other events.
Manager Lukas Wilfling said there would be a gunfire and buffet breakfast as well as full bar service from early morning.
"We will be starting the day from 6am, with bacon and eggs in the front carpark," he said.
"From 12pm, we will be holding Canberra's largest two-up competition, with all proceeds going towards Legacy."

Mercure Canberra will also have several food stalls in their front carpark, and regular dinner services from 5.30pm.
Siren Bar and Restaurant and Hopscotch, who are hosting two-up at noon, are also raising funds for Legacy.
Siren Bar and Restaurant will also have a gunfire buffet style breakfast from 6am, both the NRL and AFL on screens throughout the day, and live music in the evening.
Hopscotch will have two-up from noon to 5pm, alongside an AFL bash, live music and a DJ set from 9pm until late. Events at Hopscotch will raise money for Legacy.
Legacy ensures families of veterans are suffering no financial or social disadvantages, by providing social connections, developmental opportunities and financial aid to those in need.
