Emergency services were called to the scene after a four-wheel-drive and a sedan clipped each other whilst travelling south near the intersection with Bungendore Road shortly after 6pm.
The four-wheel-drive veered left, leaving the roadway before rolling several times.
The male driver suffered serious spinal injuries and was taken to Canberra Hospital along with two other occupants.
The sedan, carrying four occupants, veered into the middle verge of the dual carriage way. One of the passengers was also taken to Canberra Hospital with minor injuries.
The crash caused extensive delays for Canberra-bound drivers with southbound lanes expected to be closed for several hours. Traffic is now being diverted through the northbound lanes, while northbound traffic is travelling along the Old Federal Highway.
NSW Police are investigating the crash and witnesses are urged to contact Crime Stoppers on 1800 333 000.
