The car crashed into the tree on the median strip, just near the intersection with Cadell Street about 6am, and left debris on the road nearby.
Police said a passerby helped a passenger out of the car, but the driver received fatal injuries in the crash. It is understood the passenger's injuries are not life-threatening. Police are still determining whether the car caught fire on impact, or afterwards.
"ACT Fire and Rescue were at the scene and extinguished the fire. ACT Ambulance Service also attended," police said in a statement.
"ACT Policing's Collision Investigation and Reconstruction Team are investigating and will prepare a report for the ACT Coroner."
Antill Street was closed to traffic between the Phillip Avenue roundabout and Gale Street on Sunday morning, but has since been reopened.
Police said the man was the eighth person to die on ACT roads this year.
It comes just over a month after the territory's last road deaths.
