Police said an aggravated robbery had occurred at the supermarket just before 11am.
''At around 11am, police attended the Lyons shops after reports that a man had entered the IGA supermarket and threatened staff with a knife,'' a spokesman said.
''The man then fled the store with the cash register, which contained a sum of money, and was seen getting into a maroon Holden Commodore station wagon.
''A short time later, just after 1pm, police saw a vehicle matching the description in Weston.
''A traffic stop was conducted and a search of the vehicle was undertaken.''
Police took the man and a woman, both from Red Hill, into custody.
The man will be charged with aggravated robbery and the woman will be charged with aiding and abetting an aggravated robbery.
