Addressing Labor members at the ALP national conference on Friday, Mr Shorten said, "Let us make this the first decade where our head of state is one of us.
"We can be an Australian republic, with an Australian head of state."
Mr Shorten's declaration comes amid a new push within the party to restart momentum on the republic debate.
On Friday afternoon, the national conference passed a resolution that a future Labor government appoints a minister or parliamentary secretary with responsibility for promoting a republic.
In a similar process to the party's current platform, the conference also endorsed a three-step procedure to get to a republic, which includes a constitutional convention, a plebiscite to decide on a model and a referendum to change the constitution.
A co-convenor of the Labor for an Australian Republic group, Labor MP Matt Thistlethwaite, said the republican movement had learned from the 1999 referendum, "where republicans were effectively divided".
The group has identified eight possible models, which include directly electing a president, Parliament electing a president and a 50-50 model.
Mr Shorten's comments were welcomed by the Australian Republican Movement, which described them as a "game changer", representing the first time a Labor leader had set a timeframe in a generation.
But ARM national director Tim Mayfield called for a quicker timeline to a republic and said his organisation wanted to see a plebiscite by 2020 to ask Australians if they wanted a republic.
"We are a proud, multicultural, independent country today, not next year, and not next century. It's time to ask the Australian people the question again."
The republic debate has struggled since Australians voted against the idea at a referendum in 1999, under the Howard government.
Last year, polling put support for a republic at a 20-year low, with just over 39 per cent of those surveyed saying they supported a republic.
While previous recent Labor leaders Kevin Rudd and Julia Gillard were republicans, they did little to advance the cause when in office.
Before meeting the Queen in 2008, Mr Rudd said the issue was not a "top order question".
Ms Gillard said Australia should become a republic when "we see the monarch change".
