The Spanish side were eliminated from the Champions League and dropped into the second-tier continental competition having come third in their group after a disappointing campaign.
United began in the Europa League but were forced into the play-off stage after finishing second in their group, behind Real Sociedad.
The pair met in the Champions League finals of 2009 and 2011.
Barcelona manager Xavi said: "We will have to compete, going to Manchester against a really top rival and an historic side which has grown a lot with the arrival of (Erik) ten Hag.
"(They have) great individual players and it is the toughest rival once again in the Europa League."
Juventus, another giant coming down from the Champions League ranks, will face Nantes.
German sides Union Berlin and Bayer Leverkusen will play Ajax and Monaco respectively.
Other match-ups made in Monday's draw are Sporting Lisbon v Midtjylland, Shakhtar Donetsk v Rennes, Sevilla v PSV Eindhoven and Salzburg v Roma.
The Europa League knockout round play-offs were introduced last season. The eight runners-up from the competition's group stage face the third-placed teams from the Champions League group stage. The eight group winners go straight into the last 16.
There is a similar format in the third-tier Europa Conference League.
Thus after being eliminated from the Europa League, Braga and Lazio will face Fiorentina and CFR Cluj, respectively, in the Conference League play-offs.
In both competitions the first legs of the ties are scheduled for February 16, with the second legs a week later on February 23.
