The 92-year-old is engaged to retired molecular biologist Elena Zhukova, 67, according to the New York Times.
Their wedding is scheduled for June at Mr Murdoch's California vineyard and estate, the Times reported, which will mark a year since they started dating.
It's been reported Mr Murdoch met Ms Zhukova through his third wife, Wendi Deng.
Ms Zhukova left Moscow for the US in the early 1990s after splitting with her oil magnate husband Alexander Zhukov.
They had one daughter together, Dasha, who was married to Russian oligarch and former Chelsea FC owner Roman Abramovich for nine years.
She is now married to Stavros Niarchos III, the grandson of the late Greek shipping tycoon Stavros Niarchos.
The engagement news comes after Mr Murdoch's weeks-long engagement to 66-year-old Ann Lesley Smith, a former dental hygienist, radio host and prison chaplain, was called off in April 2023.
"I dreaded falling in love but I knew this would be my last. It better be. I'm happy," Mr Murdoch told the New York Post after announcing his engagement to Ms Smith.
Mr Murdoch split from his fourth wife, Jerry Hall, in August 2022 after six years of marriage.
He has six children with three of his wives.
The media mogul stepped down as leader of the Fox News' parent company and News Corp in 2023, handing over the reins to his son Lachlan.
