Emergency services were alerted to the collision in the southbound lanes of the highway, at 6.25pm on Monday night.
A NSW Police spokeswoman said emergency services responded to reports of a collision between a semi-trailer, a smaller truck and two cars in the southbound lanes of the Hume Highway.
"The male driver of the semi-trailer was ejected from his vehicle and died at the scene," the spokeswoman said.
"The drivers of the other vehicles and their passengers were assessed by paramedics on site before being taken to Goulburn Base Hospital for further treatment."
One southbound lane of the highway remained closed on Tuesday morning as traffic was directed around the crash site.
A spokeswoman for the traffic management centre said police had finalised their onsite investigations, but the closures would remain in place for the majority of Tuesday due to complex salvage operations.
She said drivers should expect some minor delays.
