The island state opened its border with the ACT on October 26 and will allow NSW residents to enter without quarantine from Friday.
The first Link Airways flights from the Nation's Capital to Hobart in seven years departs at 9.30am on Thursday. There are currently four flights weekly which Link Airways say will increase to 11 services next year.
The new route starts as Canberrans have been assured travel to Victoria will restart on November 23 as both the ACT and NSW are set unlock their border restrictions with the state which has recorded five days of no new cases.
Qantas has welcomed the announcement saying it will run 250 services between NSW and Victoria from November 23.
Western Australia will also open to the ACT from November 14 and NSW residents were given the green light to travel to Queensland without quarantine, three weeks after ACT residents were allowed into the Sunshine State.
Canberra Airport head of aviation Michael Thomson has welcomed the rolling border announcements this week which has led airlines to put on more services.
"We are delighted to see the steady return of aviation with flights consistently increasing each week as border restrictions ease across Australia and with more flights and more destinations on offer like Hobart, we are confident that this will continue," he said.
Link Airways chief executive Andrew Major said the demand for a Canberra to Hobart route had been clear before the pandemic.
"The demand for this route has been there from the beginning, so we're particularly pleased to now facilitate the route," he said.
"The new partnership between Link Airways and Canberra Airport - two Canberra based companies - has come at just the right time and we're excited to see what the future holds."
Link Airways will also operate an additional service between Canberra and Newcastle from November 20.
