The Queanbeyan location at the Riverside Plaze shopping centre will shut down on May 25.
The announcement came after Target's Batemans Bay store closed on April 20.
Staff at the Queanbeyan store were told of the planned closure in February, with notices about the closure placed outside the store in recent days.
A Target spokesman said it was working with staff ahead of the shut down date.
"Target is working hard to ensure our Queanbeyan team members can join another Target team at a store convenient to them, or across the wider Westfarmers retail businesses," the spokesman said.
"Target wants to thank our Queanbeyan team, the local community and all of our customers for shopping with us."
The spokesman did not confirm how many staff would be affected by next month's shut down.
The retailer announced last year it would close dozens of stores as part of an effort to reduce its footprint by 20 per cent over five years.
At the time of the announcement last June, there were 305 stores across Australia.
Westfarmers' chief executive Guy Rosso said last year Target was aiming to shift away as a traditional rival to Kmart and Big W and look to be a more fashionable brand to rival fast-fashion chains H&M and Zara.
There are four Target stores in the ACT, with locations at Canberra Centre, South Point shopping centre, Westfield Belconnen and Cooleman Court.
