Roads ACT Director Tony Gill traffic signs and controllers would be in place this week, with partial lane closures expected.
But he said preliminary work on the project was not expected to cause traffic delays.
“The ACT Government thanks motorists and other road users for their patience during this work,” he said.
The new 11.5-kilometre, $288 million four lane road will link the Federal Highway with the Monaro Highway.
It is intended to enable better traffic movement from the north to the southside of Canberra and provide better access to the Majura Valley, and is due to be completed in 2016.
It will have a 100km/hour speed limit and is expected to carry 30,000 more vehicles a day by 2031 than Majura Road, which now links the highways.
Mr Gill said from Wednesday, controllers would be directing traffic south of the alignment on Morshead Drive, Fairbairn Avenue and Monaro Highway, near Canberra Airport.
They would also be working at the Federal Highway from Monday 21 January.
